Does the sony xperia U support usb on the go (for pen drives) ? and if it does, do we need to root the phone to use the functionality or it can be used normally just by connecting a otg cable and using some file explorer app ?

Yes, the phone support USB OTG.
BUT
Most pen drives demand more power than the phone can supply meaning it will not work.
The USB OTG feature is mainly so you can connect mouse, keyboard and gaming controller to the phone, preferably using LiveDock which enables all this.
So - there's no harm in trying but don't expect this to work.
